New strategy in the discovery of natural antituberculosis agents from French Guiana flora

Résumé

Plants have been used for centuries in traditional medicine to treat various affections, and are widely studied by chemists for the molecular diversity they offer. Ethnopharmacology aims to valorize traditional knowledge on plants, as they represent an important source of new compounds of therapeutic interest. However, it can be challenging to describe the metabolites responsible for the activity in a sample as complex as a plant extract. The recent use of the MS-based Molecular Network approach can facilitate structural dereplication and accelerate the annotation of new structural entities in complex samples. Combining the most recent metabolomics tools with a process inspired by high troughput screening will allow to annotate large datasets, and get rapid and accurate results on the metabolites reponsible for the studied therapeutic properties. This poster aims to present a workflow for the research of new antituberculosis agents in plants, using metabolomics combined with an ethnopharmacological approach. This study focuses on French Guiana, as it is the French department with the highest incidence of tuberculosis. Plants selection led to 12 species collected in French Guiana, and used traditionally by local populations to treat tuberculosis symptoms. Metabolomics data were aquired using UHPLC-HRMS and GC-MS. The results of bioassays carried on Mycobacterium tuberculosis, combined with the aquired data enable the construction of a molecular network, which will accurately highlight metabolites presenting antituberculosis properties. Mixtures NMR analysis will then confirm the proposed annotations. Through this workflow, the Guiana’s flora becomes a chemical library, the goal being to emphasize its antituberculosis potential.
